I have a focus Thron2 and I have really low space to fit a bigger chainring, actually I have a 34.
I’m trying to figure out if I can simply use direct mount and go with e-thirteen 36 chainring or should I go with a spider 104 and apply chainring on it. I’ve tried to search on internet info about chainline of the boost spider version but i haven’t found anything...
I need 52/53 boost chainline.

It can be done, using a spider and 104bcd chainring, you might find this thread useful

What exists? Bosch Gen4 + direct mount? SRAM or Shimano . 12 speed? Eagle?

Assuming you have a boost rear hub ( 148mm) your existing chainwheel set up with be 3mm offset ( 52mm). It is possible to get 6mm offset for Bosch Gen ( which is ISIS) , which is designed for super boost rear hub and that may povide the chainstay clearance you need but at the expense of a more extreme chainline in the lower gears on a normal boost hub.

I would think it would be OK for the highest 8 or 9 gears.
